MOTS-C is unusual: it is one of a small group of peptides encoded not in the cell nucleus but in mitochondrial DNA. Discovered at the University of Southern California, it is studied as a signal from mitochondria to the rest of the cell about metabolic state, and it has become a reference compound in metabolism and exercise-physiology research.
Lee et al. (2015, Cell Metab) reported that MOTS-C acted on skeletal muscle through the folate cycle and AMPK, improving insulin sensitivity and preventing diet-induced obesity in mice.
Reynolds et al. (2021, Nat Commun) reported that MOTS-C improved physical performance in young, middle-aged and old mice and that its levels rose with exercise in human muscle.
