Amylin is a hormone released alongside insulin that signals satiety and slows gastric emptying. Natural amylin aggregates and is short-lived, so cagrilintide was engineered with a fatty-acid chain and stabilizing substitutions to last long enough for once-weekly study designs. It is best known for research in combination with GLP-1 agonists.

Phase 2 data published in The Lancet (Frias et al., 2023) and later phase 3 programs examined cagrilintide combined with semaglutide, reporting larger body-weight reductions than either component alone.
