Ipamorelin is a synthetic five-amino-acid peptide that activates the ghrelin receptor, one of the two main switches that trigger growth hormone (GH) release from the pituitary. It belongs to the same family as GHRP-6 and GHRP-2, but was developed specifically for selectivity.
Raun et al. (1998, Eur J Endocrinol) described Ipamorelin as the first selective GH secretagogue: in animal models it released GH with potency comparable to GHRP-6 without significant increases in ACTH or cortisol, even at much higher doses.
Because it acts on the ghrelin pathway, Ipamorelin is often studied with GHRH analogues such as CJC-1295, which act on the other pathway. Stimulating both receptors produces a larger GH response in research models than either alone.
