IGF-1 vs IGF-1 LR3: The Difference, Why Labs Use LR3, and the Risks

Short answer: IGF-1 is the natural hormone; IGF-1 LR3 is an engineered version that is 13 amino acids longer and has one amino acid swapped. Those two changes stop it from being captured by the binding proteins that normally hold IGF-1 in check, so it stays active far longer and is much more potent in cell culture. That is why it is a standard reagent in cell biology labs, and why its risks in the body are a step up from natural IGF-1. The closest approved drug, mecasermin (natural IGF-1), carries warnings for low blood sugar, which affected 42% of patients in its trials.

Why binding proteins matter

In the blood, nearly all IGF-1 is bound to six IGF binding proteins (IGFBPs), mostly IGFBP-3. Bound IGF-1 is inactive; the binding proteins act as a reservoir and a brake, releasing small amounts where needed. The “R3” change (arginine at position 3) and the long extension both weaken this binding. Free from its brake, LR3 hits the IGF-1 receptor harder and for longer. In cell culture, that is exactly the point.

Why labs use LR3

IGF-1 LR3 was developed in Australia in the early 1990s as a cell-culture supplement. Serum-free culture media need a growth signal, and natural IGF-1 gets mopped up by binding proteins the cells themselves secrete. LR3 keeps working, so it is widely used in biomanufacturing and in muscle, metabolism and stem-cell research.

Risks: what the approved IGF-1 drug teaches

There are no human trials of IGF-1 LR3. The best guide to what IGF-1 receptor activation does in people is the label of mecasermin (natural IGF-1), which has a long clinical record:

  • Hypoglycemia: 42% of children in trials had at least one episode; severe cases caused seizures. IGF-1 lowers blood sugar much like insulin.
  • Lymphoid tissue growth: tonsil enlargement in 15%.
  • Intracranial hypertension, allergic reactions including anaphylaxis, and skeletal effects.
  • Malignancy: the label warns of postmarketing reports of malignant tumours and says to stop treatment if one develops.

Why this matters for LR3: LR3 escapes the binding proteins that limit natural IGF-1, so its effect on blood sugar and cell growth is expected to be stronger and harder to control. Low blood sugar is the most immediate danger.

Frequently asked questions

Cliquez sur une question pour afficher la réponse.

What is the difference between IGF-1 and IGF-1 LR3?

IGF-1 LR3 has a 13-amino-acid extension and an arginine in place of glutamic acid at position 3. These changes greatly reduce binding to IGF binding proteins, making it longer-acting and more potent than natural IGF-1.

Why is IGF-1 LR3 used in labs?

It stays active in serum-free cell culture because it is not captured by binding proteins that cells secrete, so it is a common growth supplement in cell biology and biomanufacturing.

What are the risks of IGF-1?

The approved IGF-1 drug mecasermin lists hypoglycemia (42% in trials), tonsil enlargement, intracranial hypertension, allergic reactions and reports of malignant tumours. LR3 is expected to act more strongly.

Is IGF-1 LR3 approved?

No. Only natural IGF-1 (mecasermin) is approved, for severe primary IGF-1 deficiency in children.

Is IGF-1 LR3 banned in sport?

Yes. WADA prohibits IGF-1 and its analogues under S2.
