Selank vs Semax: Same Lab, Two Very Different Peptides

Same lab, different missions: Selank and Semax were both developed at the Institute of Molecular Genetics in Moscow, both end in the same stabilizing Pro-Gly-Pro tail, and both get lumped together as “nootropic peptides.” Yet they come from completely different parent molecules and have been studied for different things. Here is how they actually differ, what the research says about each, and why some labs study them side by side.

The shared trick: Pro-Gly-Pro

Short natural peptides are usually destroyed by enzymes within minutes. The Moscow group’s solution was to attach a three-residue tail, Pro-Gly-Pro, to the active fragment. Proline-rich sequences resist many common peptidases, so the tail extends how long the molecule survives in research models. That design choice is the only thing the two compounds really share.

Selank: the calm one

Selank starts from tuftsin, a four-amino-acid immune-modulating peptide (Thr-Lys-Pro-Arg) that occurs naturally as a fragment of an antibody. Adding Pro-Gly-Pro produced a molecule that, in rodent studies, showed anxiolytic-like behaviour without the sedation, muscle relaxation or memory effects associated with benzodiazepines.

Proposed mechanisms include modulation of GABAergic signalling and changes in serotonin and dopamine metabolism, alongside effects on immune cytokines inherited from tuftsin. Gene-expression studies from the same institute reported shifts in genes related to neurotransmission and inflammation.

Semax: the focus one

Semax starts from a fragment of adrenocorticotropic hormone (ACTH), the pituitary hormone that tells the adrenal glands to make cortisol. The chosen fragment, ACTH(4-7), carries neuroactive properties but not the hormonal action on the adrenals, so Semax is studied for brain effects without driving cortisol release.

The most cited finding comes from Dolotov and colleagues (2006, Brain Research), who reported that Semax increased levels of BDNF (brain-derived neurotrophic factor) and activation of its receptor TrkB in the rat hippocampus. BDNF supports the survival and plasticity of neurons, which is why Semax appears in learning, memory and ischemia models. Other studies examined gene-expression responses after experimental stroke in rats.

How strong is the evidence?

Honestly: moderate and concentrated. Most published work comes from Russian institutions, much of it in Russian-language journals, and both compounds are registered as medicines in Russia but have not gone through Western regulatory trials. Independent replication outside that research network is limited. That does not make the findings wrong, but it is the context any serious research design should account for.

Bottom line: both peptides are interesting tools for studying stress and cognition pathways. Neither is an approved treatment in Canada, and research findings in rodents are not evidence of effects in people.

How researchers choose

  • Stress, anxiety-like behaviour or immune-neural crosstalk? Selank is the closer fit.
  • Neurotrophins, learning models or ischemia? Semax has the more relevant literature.
  • Comparing two Pro-Gly-Pro peptides from different parents? Running both side by side isolates the effect of the active fragment, since the stabilizing tail is identical.

Other neuropeptides in the same research space include DSIP (sleep models) and Pinealon (neuroprotection models).

Frequently asked questions

Cliquez sur une question pour afficher la réponse.

What is the difference between Selank and Semax?

Selank is derived from the immune peptide tuftsin and studied mainly in anxiety and stress models. Semax is derived from an ACTH fragment and studied mainly for BDNF, cognition and neuroprotection.

Why do Selank and Semax both end in Pro-Gly-Pro?

The Pro-Gly-Pro tail resists enzymatic breakdown and was added by the same Moscow institute to extend each peptide’s stability.

Does Semax affect cortisol?

Semax uses an ACTH fragment that does not carry ACTH’s hormonal action on the adrenal glands.

Are Selank and Semax approved in Canada?

No. Both are registered in Russia but not approved by Health Canada. They are sold in Canada as research chemicals for laboratory use only.
