PT-141 is a cyclic peptide derived from Melanotan II. During MT2 research, investigators noticed effects mediated by the MC4 receptor in the brain, and PT-141 was developed to study that pathway. It later became the active ingredient of an approved prescription drug in the United States, which makes it one of the few research peptides with a full clinical program behind it.
PT-141 acts in the central nervous system rather than on the vascular system, which distinguishes its mechanism from PDE5 inhibitors in research models.
Phase 3 RECONNECT trials led to regulatory approval in the United States in 2019 for a specific indication.
